Always plan ahead when you receive Roses. When received, cut the stems and place the Roses in water but do not remove the protective cardboard and sleeve of Rose Dark X-pression for at least 4 hours. This is the optimal way for the stems to draw water. Do not process flowers close to air vents, next to fruit (which causes ethylene), or in freezing temperatures. After the Roses are processed, carefully remove the cardboard and position Roses in a vase. If needed, carefully remove a few guard petals. For best result, always cut the stems in an angle. Remove excess foliage to prevent leaves from touching the water. For maximum enjoyment, keep Rose Dark X-pression away from the sun (windows) and drafts (HVAC vents). They were beckoning mountains with a brown grass love.

The Santa Lucias stood up against the sky to the west and kept the valley from the open sea, and they were dark and brooding — unfriendly and dangerous.
And the western mountains of Santa Lucia, separating the valley from the ocean, stood against the background of the sky as a dark gloomy ridge – these mountains were inhospitable, formidable.
I always found in myself a dread of west and a love of east.
That which lay to the west struck fear in me, but I dearly loved everything that stretched to the east.
Where I ever got such an idea I cannot say, unless it could be that the morning came over the peaks of the Gabilans and the night drifted back from the ridges of the Santa Lucias.
I can explain such a strange bias, perhaps, only by the fact that the morning descended into the valley from the peaks of Gabilan, and the night crept from the ledges of Santa Lucia.
It may be that the birth and death of the day had some part in my feeling about the two ranges of mountains.
I saw where the day is born and where it dies – maybe this is the reason for the different attitude to the two mountain ranges.
From both sides of the valley little streams slipped out of the hill canyons and fell into the bed of the Salinas River.
In the winter of wet years the streams ran full-freshet, and they swelled the river until sometimes it raged and boiled, bank full, and then it was a destroyer.
On both sides of the valley, streams flowed into the river from the canyons, In rainy years they turned into fast, deep streams in winter, and the river swelled so much that sometimes, not fitting in the banks, it boiled angrily and began to rage – its rage then was crushing.
The river tore the edges of the farm lands and washed whole acres down; it toppled barns and houses into itself, to go floating and bobbing away.
The river gnawed at the edges of the estates and flooded the fields; she overturned houses, sheds, and they floated away, heeling and trembling on the waves.
It trapped cows and pigs and sheep and drowned them in its muddy brown water and carried them to the sea.

In Vivo Targeted Expression of Optogenetic Proteins Using Silk / AAV Films
The use of silk / AAV to express optogentic proteins overcomes the limitations of current approaches. Although many studies have successfully used AAV injections to express optogenetic proteins, it is difficult to align the expression of the tip of the optical fibers, the regions around the length of the conical fibers, and to the viewing area of the lens with a smile.Due to the mismatch of optical components and optogenetic expression, stereotactic injections may be unreliable and not a lot of experimentation. The silk / AAV labeling method we described here solves this problem. It also simplifies the procedure by eliminating a second surgical step and, in some cases, eliminating the need for a second surgery. It can also be difficult to use to obtain a broad expression under the cranial windows of viruses, and the experimenter usually performs lengthy operations to inject the virus into multiple locations. The ability to obtain wider expression over large cortical regions by simply covering the cranial windows with silk / AAV is simplified, eliminating the need for many invasive injections.
Another potential advantage of the silk / AAV method is that it can induce less inflammation in the nerve tissue, compared to viral injection. Injecting a high titer of AAV into the brain may induce inflammatory reactions such as reactive astrocytosis, which have the potential to alter cellular and distribution patterns 14 , 15 (although usually such potential complications are ignored).Silk films induce little immunogenicity response on their own 13 and silk / AAV films are expected to release a virus over many hours or days 16 , which can reduce viral load in surrounding tissues and reduce immunogenicity responses. With traditional approaches in which device implantation is preceded by AAV injection, inflammatory reactions can arise from implantation and injection. In the future, it will be desirable to systematically compare traditional approaches and the silk / AAV method to determine whether silk / AAV films reduce overall inflammatory responses.
Several steps are critical to the successful use of silk / AAV films. Most importantly, the coating of the optical fibers should be done carefully as described in the methods and the location of the dried films should be assessed carefully by visual inspection under a microscope to ensure compact, films in the right place, and adhere to the face of the optical fiber. Any silk / AAV on the sides of the fiber optic will result in an expression of interest outside the region, and during implantation and lead to unreliable or expression may not break producing films that protrude beyond the face of the fiber.The methods that we describe for the application of silk / AAV implantable devices can be tailored to the use of any materials that are readily available and permit accurate deposition of small volumes of silk / AAV.
A little practice is required to ensure accurate and reproducible results. If expression is observed along the grain direction, it is likely that the silk film is dried on the fiber side, instead of the fiber face. Repeat the manufacturing process and carefully inspect the dried implants for marks that are drying films on the fiber side.Because silk / AAV films are optically transparent, it can help in the practice of applying silk mixed with dye (fast green or dye) to better visualize the shape of the resulting films ( Fig. 2 c ). If expression fails, it is likely that the silk film was knocked out of the fiber tip during implantation. We suggest using stocks of viral titer when making implants. For optical fibers, this reduces the overall volume that must be applied to small diameter fibers.If coating size is a concern, wait longer between each 10 nL application for complete drying of the storage drops. Silk / AAV droplets dry faster under a warm lamp. For cranial windows, a high viral titer may be necessary to provide adequate viral load via Pia or Dura. Some types of implants can dissolve the silk and release the AAV more easily than others. We found that cranial windows implanted above the brain surface require a lower silk / virus ratio to achieve robust expression, possibly due to different cerebral spinal fluid dynamics or protease activity.If expression cannot be increased by increasing the effective concentration of AAV, decreasing the volume of hydrous silk is a plausible alternative.
Finally, it is important to store the optical components properly and implant them fairly soon after they are prepared. We have shown that coated fibers that are refrigerated under vacuum can be stored for many days prior to use. Vacuum storage removes residual moisture 17 , which can reduce the solubility of silk films, and also help maintain viral efficacy.Ideally, optical fibers should be implanted within 24 hours of manufacture. However, we find that silk / AAV-coated fibers are stored under vacuum drive with similar expression levels when implanted 7 days after fabrication ( Fig. 4B ). In contrast, the cranial windows and lenses of the smile have forced the most reliable expression when they are dried at room temperature and used within hours of preparation. The reason for this inequality remains unclear.Further research may be necessary to clarify the preparation and storage conditions to extend the shelf life.
Silk / AAV-coated cranial windows have significant potential because they drastically reduce surgical times and are extremely easy to manufacture, but currently this method has limitations. Covered cranial windows evenly label large areas of the cortex and drive sufficient expression in layer 2/3 for GCaMP imaging, with slightly less expression in deeper layers.However, stereotaxic disc injections are more reliable in expression and provide more control over the layers intended for expression. A reliable expression was only achieved when Dura was removed. Although for many 2-photon imaging experiments Dura is often removed to improve image quality 8 , for many experiments it is desirable to obtain markings in a less invasive manner. Therefore, we examined our ability to use silk / AAV to mark cortical regions without removing Dura.We received some markings, but it is possible that this is a consequence of Dura’s injury during the preparation of the craniotomy. Further research is needed to ensure that the coated cranial windows should be used to reliably label the cortex without removing Dura.
The preparation of aqueous fibroin silk from silkworm cocoons is detailed in Rockwood et al. (2011) 18 . Aqueous silk fibroin is currently commercially available (5% w / v). Although most of our experiments were performed using stocks of aqueous silk fibroin prepared in our laboratory (5-7.5% w / v), we obtained similar results using commercial aqueous fibroin.Aqueous Fibroin is stabilized at 4 ° C for up to 3 months, after which it spontaneously passes out of the liquid hydrogel 18 . We recommend dividing ~ 1 ml Aligote Fibroin stocks and stored at -80 ° C. 1 ml Working Aligote (enough to cover hundreds of implants) can be stored at 4 ° C and used until it starts to gel. Be careful not to shake, vortex, agitate, or aggressively Pipette an aqueous Fibroin, shearing forces can lead to gelation 19 , 20 .
Silk / AAV films allow a wide range of expression patterns, from widespread cortical expression under the cranial windows, to precise subcortical expression at the tip of small diameter optical fibers. These techniques were developed to take advantage of common AAV expression vectors, but can most likely be used to disperse other expression vectors like Lentiviruses or rabies viruses into the brain. Silk films can also be fabricated into three-dimensional shapes to improve viral release into tissue.For example, in order to drive strong expression under cortical windows without the use of a durotomy, the cranial windows can be coated with arrays of microneedle silk, which would pierce the fool and release the virus into the deeper layers of the cortex 21 . Further refinement is likely to lead to improved properties of the virus release, and new applications for silk / AAV films.

In debates and discussions of politicians, social inequality is given less attention than property inequality. However, according to the 2019 BCG SEDA analysis., the correlation between social equality and well-being is higher than between income equality and well-being. Government dashboard and its capabilities
As the report notes, creating comprehensive scorecards can help governments capture important signals, including those of social inequality.Many countries, such as New Zealand and the United Kingdom, are already making active efforts to go beyond purely economic indicators (such as GDP) and focus on the overall well-being of their citizens when making policy and budgeting. The next step could be the creation of a dashboard that displays a complete picture of the situation in the country. It should include real GDP per capita growth, objective measures of well-being as reflected in the SEDA and other similar rankings, and more subjective measures such as overall happiness.
As shown in the report, this “three-dimensional” scorecard reveals problems that are easily overlooked when evaluating just one score. For example, a country’s score in the UN World Happiness Report usually matches its level of well-being according to SEDA’s wealth-to-wealth ratio. The coefficient assesses the extent to which a country manages to “convert” its wealth into the well-being of its citizens. However, there are many countries with relatively high scores on this SEDA score, while their happiness levels are lower than expected.It is only by looking at these two indicators together that governments will be able to identify alarming biases – and the drivers behind them.
“When a government focuses on just one metric, such as GDP, it runs the risk of missing important signals that indicate problems in the country,” notes Enrique Rueda-Sabater, senior advisor at BCG and co-author of the report. “The three-dimensional approach we are proposing allows us to get a clear picture and understand in which areas action needs to be taken.”
Using indicators from open sources, we assess the country for each dimension. In total, the assessment is based on 40 indicators, the values of which are determined based on the latest available data. (The 2019 analysis mainly used 2017 data – note that recent changes may not be reflected in the analysis.) The value of each indicator is normalized on a scale from 0 (lowest score among 143 countries) to 100 (highest score).Based on these normalized indicators, a score is calculated for each of the ten dimensions. We can use these scores to analyze well-being in three dimensions:
1. SEDA score
We aggregate scores across 10 SEDA dimensions to give a single score for a country. It can be used to compare a country to other countries or groups of countries. In general, richer countries tend to score higher. The ten SEDA dimensions also help prioritize action, as comparing a country to other countries or groups of comparable countries reveals key strengths and weaknesses.Armed with this data, governments can begin to define strategies to address the most pressing issues.
2. Changes in the SEDA score
With a 12-year SEDA benchmark database, we can track the change in score over that period. We can also track changes for each dimension of the SEDA score.
3. Wealth-to-wealth ratio
Based on SEDA estimates, we can investigate how successful countries are in converting their wealth (per capita income) into wealth.To do this, we use the ratio of wealth and well-being. The ratio compares a country’s actual SEDA estimate with the estimate that would be expected given its gross national income per capita. Thus, the ratio is a relative measure of the “conversion” of a country’s wealth to the well-being of its population. Countries with a score of 1.0 achieve a level of well-being that is consistent with that expected at their income level. Countries with a score above 1.0 have achieved higher levels of well-being than expected at their gross national income, while countries with a score below 1.0 have below-expected well-being..
